O Tony Award de Melhor Atriz em Musical (em inglês: Best Performance by a Leading Actress in a Musical) é um dos prêmios oferecidos no Tony Awards, cerimônia instituída em 1947 como Antoinette Perry Awards for Excellence in Theatre, entregue em honra às atrizes que se destacam em papéis principais de qualidade em peças musicais, seja uma nova produção ou uma remontagem, em determinado ano. O prêmio leva o nome de Antoinette Perry, atriz americana falecida em 1946.

Honrarias em diversas categorias são apresentadas na cerimônia anualmente pela Tony Award Productions, uma joint venture da The Broadway League e da American Theatre Wing, para "homenagear as melhores performances e produções teatrais do ano anterior."[1]

O prêmio foi originalmente chamado de Prêmio Tony para Atrizes - Musicais. Grace Hartman foi a primeira vencedora no 2º Tony Awards por sua interpretação de vários personagens em Angel in the Wings. Antes de 1956, os nomes das indicadas não eram tornados públicos;[2] a mudança foi feita pelo comitê de premiação para "ter um impacto maior nos espectadores do teatro".[3]

Em 1965, Liza Minnelli, aos 19 anos, tornou-se a atriz mais jovem a ganhar o prêmio. Ela foi seguida por Lea Salonga, aos 20 anos, em 1991. Em 2017, Bette Midler, aos 71 anos, tornou-se a atriz mais velha a ganhar o prêmio. Ela foi seguida por Victoria Clark, aos 63 anos, em 2023.

Angela Lansbury detém o recorde de artista com mais vitórias nesta categoria, com um total de quatro. Chita Rivera tem o maior número de indicações, com oito. As personagens Rose em Gypsy e Anna Leonowens em The King and I detêm atualmente o recorde de mais premiadas, cada uma com três vitórias. Rose também detém o recorde de mais indicações, com seis.
